How to make slides.　スライドの作り方。今日もせっせとスライドづくり。

UVレジンコーティング液を使った珪藻のスライドの作り方の説明。

Here is my current method of making the prepart. For your reference.

First, I prepared a sticker with holes to be attached to the glass slide.

It is used as a spacer to prevent the cover glass from slanting.

Follow the guide to apply the sticker. Seal thickness is 0.05mm (50μm) diameter 14.5mm

Next, prepare the circular cover glass 15 mm (a bit high), UV resin coating solution and a painting stick.

Apply to 15φ cover glass. As thin as possible.

Then arrange them under a stereomicroscope. (Not pictured)

Even though it is thinly coated, it looks like a puddle under the microscope. Submerge the diatom in the puddle of resin solution with the tip of your eyelash. The disadvantage of this UV resin solution is that the diatom will flow if you move it a little. It would be better if it had a little more viscosity, but if the viscosity is too high, the air bubbles will not escape.

Once lined up, they are hardened with a UV lamp. A few dozen seconds.

After lightly hardening, add a drop of UV resin coating solution on top. (Make sure there are no air bubbles).

A drop is also placed in the middle of the sticker on the glass slide.

Finally, it's time to paste.

Attach the liquid dripped on the cover glass and the liquid dripped on the slide.

Align well. Since the hole seal is slightly small, once the liquid has been applied to the entire circumference, solidify it with a UV lamp to complete the process.
